The Study on Siderophore Biosynthetic System in Pseudomonas Sp. PY002



Abstract
In iron-limited environment, strain PY002 produced yellow-green fluorescent pigment. The absorption spetra of a culture supernatant showed two peaks at 280 nm and 400 nm. Yellow-green pigment production was enhanced at 30¡Ébut repressed at
37¡Éand
41¡É.
To identify siderophore-producing clone, siderophore biosynthesis defective mutant MP114, was complemented by using triparental conjugation method. From the complementation exprements, pCOM61 was able to exhibit Flu+/Sid+ phenotype. The
restriction
map
of insert DNA from the pCOM61 was constructed using the restriction enzymes such as EcoR¥°, Hind¥²and Xho¥°. By subcloning, it was revealed that EcoR¥°-EcoR¥°fragment (7 kb) of pCOM61 chould complement the MP114.
